INTRODUCTION
Rapid Brucellosis antigen rapid test kit use rapid immunochromatographic test to detection brucellosis antigens. After the sample to be added to the loading hole, it moves along the chromatographic membrane together with the colloidal gold labeled anti-Brucellosis monoclonal antibody. When a Brucellosis antigen is present in the sample, it shows a red-wine color by binding to the antibody on the assay line. No color reaction occurs if no Brucellosis antigen is present in the sample.

TEST PROCEDURE
1. Prepare specimen
A. Pure culture bacteria: Pick up a single colony of suspected Brucella to assay buffer, cover and shake the tube vigorously for more than 30 seconds, then let it stand for 5 minutes. After the large particles settle, absorb the supernatant as the sample extraction.
B. Clinical specimens: Use swab dipped in suspected brucella in inguinal lymph node acupuncture aspirates, vaginal discharge, and diseased tissue homogenates. Immediately insert the swab into the buffer tube and agitate it at least 10 times. Then cover the buffer lid and shake it over 30 seconds. Then stand for 5 minutes until the large particles precipitated to absorb the supernatant as sample extraction.
C. Milk: Collect 20ml raw milk and centrifuge for 15 minutes at 2000g. Discard the supernatant and dip the sediment in the centrifuge with a swab. Then Immediately insert the swab into the buffer tube and agitate it at least 10 times. Then cover the buffer lid and shake it over 30 seconds. Then stand for 5 minutes until the large particles precipitated to absorb the supernatant as sample extraction.
2. Take out the test device from the foil pouch and place it horizontally.
3. Add 5 drops of sample extraction into the sample well.
4. Interpret the result in 15-20 minutes. Result after 20 minutes is considered as invalid.
INTERPRETATION OF RESULTS
Positive: Two lines appear, no matter T line is clear or vague.
Negative: Only C line appears.
Invalid: No line appears or only T line appears.
PRECAUTIONS
1. Do not use expired test or the test with damaged foil pouch.
2. The test must be used within 1 hour after opening the foil pouch.
3. The test sample should be tested immediately to avoid long time storage. When adding sample into the sample well also need to avoid bubbles.
4. Do not reuse the test kit and do not mix components from different lot numbers.
5. All specimens have potential infection. It must be strictly treated according to the rules by local states.
LIMITATION
Pawiser Rapid Brucella Ag Test kit is for in vitro veterinary diagnosis use only. All results should be considered with other clinical information available from veterinarian. For an accurate result, it is suggested to apply other method such as PCR for final determination in practice.
